Can a 40-pole front connector replace a 20-pole one?

No. The connector must match the pin count of the module. Using the wrong one will not fit or will leave channels unconnected.

What does a TOP connect module do?

It connects a signal module to field wiring through terminals, so each channel is wired on a DIN rail terminal block, not on the module.
